Isolation of Uridine Diphosphate N-Acetylglucosamine-6- phospho-1-galactose from Hen Oviduct
نویسنده
چکیده
The isolation and identification of uridine diphosphate N-acetylglucosamine 6-phosphate from extracts of hen oviduct was first reported by Strominger in 1955 (1). Since that time, essentially no information concerning the metabolic role of this compound has become available. Recently, however, brief reports from two independent laboratories (2, 3) have described the isolation of a modified compound in which the terminal phosphate of the hexosamine is found in phosphodiester linkage to the anomeric carbon of galactose. The possibility must be considered, therefore, that the original compound may represent a partial degradation product of the more highly substituted nucleotide conjugate. The present paper provides a detailed description of the isolation procedure as well as further documentation in support of the proposed structure of the galactose-containing nucleotide (Fig. 1).

While the yolk traverses the hen oviduct, albumen, shell membranes, and the calcified shell are successively secreted (3). The isthmus, a short section 2 to 8 cm in length, synthesizes and secretes the inner shell membrane, a lace-like envelope composed of fibers of an acid glycoprotein (see below). A number of nucleotides have been shown to occur in the isthmus, including cytosine diphosphate ...

It has been shown previously (1) that mung bean seedlings (Phaseolus aureus) contain uridine diphosphate derivatives of n-glucose, n-galactose, n-xylose, and L-arabinose. These seedlings, as well as a number of other higher plants, also contain enzymatic systems capable of catalyzing the reversible formation of sugar nucleotides from UTPI and various monosaccharide l-phosphates (2, 3) according...

A nucleotide isolated from a rough mutant of Salmonella typhimurium LT2 has been identified as uridine 5'-diphosphate L-rhamnose on the basis of chemical, enzymatic, and chromatographic analysis. Other nucleotide-linked sugars were isolated from this organism and tentatively identified as cytidine diphosphate abequose, cytidine diphosphate glucose, uridine diphosphate glucose, uridine diphospha...
